The authors have previously followed traditional Chinese acupuncture methods in the treatment of recurrent herpes simplex infection and have had moderately good results. However, following the successful treatment of a very severely affected patient who had suffered more than 15 recurrences per year, an alternative regime was instituted. This involved body acupuncture to immunomodulating points, direct irradiation of the herpetic lesions with He-Ne laser, daily application of aloe vera extract, and specific dietary restrictions. Using this regime they report complete recovery, with no herpetic recurrence for more than a year, in up to 83% of their patients. They also measured immunological parameters before and after treatment, finding that : serum IgG, C3 complement fraction, and the protease inhibitors ocl-antitripsin and oc2-macroglobulin all increase, while secretory IgA and C3 complement fraction in urine normalise. These preliminary results have encouraged continuation of the clinical trials of this method.

Objective: to obtain the most excellent dispending proportion of Chinese drugs for killing herpes simplex virus type- II. Methods: The antiviral effects of Flos Lonicerae and Radix Astragali which were used singly or in combination on HSV-2333 strain were observed by plaque reduction assay, compared with ACV positive control group. The antiviral effect analysis was made by Median Effect Principle. Results: In Hep- II cell system,the ED50 of directly virulicidal activity, the inhibiting effects on the multiplication and the anti-infectious action of ACV on HSV-2333 strain in the ACV group were 16.45 µg/ml,10. 85 µg/ml, 18.62 µg/ml respectively, while those in the observation group in the combination ratio of 1 (ED50) Flos Lonicerae/1 (ED50) Radix Astragali 0. 67 µg/ml, 0. 80 µg/ml, 1. 19 µg/ml. The antiviral effects of observation group were higher than that of the ACV group and the average combination index (CI) is lower, thereby the synergism of combination of Flos Lonicerae and Radix Astracali was evident. Conclusion: The combination ratio of 1 Flos Lonicerae/1 Radix Astragali is the most excellent ratio.

In order to understand the effect of "Life-Spring Liquid Extract" in resisting herpes simplex virus II (HSV-2), experimental observation in vitro was conducted by MTT and CPE inhibition, with acyclic guanosine (ACV) as the control drug. The results showed that TC50 was 1.39mg/ml, IC50 was 0.041mg/ml and treatment index (TI) was 33.9 in "Life-Spring Liquid Extract" by MTT, and the TC50 was 1.61mg/ml, IC50 was 0.055mg/ml and TI<2 9.37 in "LIFE6SPRING LIQU-SPRING LIQUID EXTRACT3 BY CPE inhibition. In comparison with ACV, "Life-Spring Liquid Extract" has remarkable effect to inhibit HSV-2 in vitro.

Kakkon-to is one of the representative traditional herb medicines (Kampo formulae) and has been used historically for the treatment of infectious diseases in China and Japan. The efficacy of this preparation was characterised using a cutaneous herpes simplex virus type 1 (HSV-1) infection in mice as a model for human viral infection. Kakkon-to at a dose corresponding to human use reduced significantly the mortality of HSV-1-infected mice and localised skin lesions. Delayed type hypersensitivity (DTH) response to HSV-1 antigen was significantly stronger in treated mice than in untreated mice. However, no histopathological difference was noted in the skin lesions between treated and untreated mice except for the size of the lesions. Kakkon-to did not inhibit the growth of HSV-1 in vitro. Natural killer cell activity, natural cytotoxic killer cell activity, and the population of T-cell subsets in spleen cells of infected mice were not affected by the drug. Kakkon-to did not augment interferon induction and anti-HSV-1 antibody production, nor increased cytokine levels such as interleukin-1 alpha, interleukin-2, interferon-gamma, and tumour necrosis factor-alpha in sera of infected mice. Thus, Kakkon-to induced strong DTH to HSV-1 in infected mice, which may have caused localisation of skin lesions and reduction in the mortality of treated mice.

The protective effect of Kanzo-bushi-to (TJS-038) was investigated on the opportunistic infection of herpes simplex virus type 1 (HSV) in thermally injured mice (TI-Mice). We have previously reported that TI-Mice were approximately 100 times more susceptible to HSV infection than normal mice (N-Mice) and that CD8+ suppressor T (ST)-cells induced by burn injury were involved in causing this increased susceptibility of TI-Mice. Increased susceptibility of TI-Mice to the infection was reversed to the levels observed in N-Mice when TI-Mice were treated intraperitoneally with TJS-038 at a dose of 5 mg/kg 1 and 4 days after thermal injury. The activity of ST-cells was greatly decreased in TI-Mice treated with TJS-038. The generation of Vicia villosa lectin-adherent CD4+ CD28+ TCR-alpha/beta+ contrasuppressor T (Contra-ST)-cells associated with the appearance of ST-cells was expanded and occurred earlier in spleens of TJS-038-treated TI-Mice as compared with that of untreated TI-Mice. The improved resistance of TJS-038-treated TI-Mice to the infection was transferred to untreated TI-Mice by adoptive transfer of Contra-ST-cells prepared from TJS-038-treated TI-Mice. These results suggest that TJS-038 may restore the resistance of TI-Mice to the HSV infection through the expanded generation of Contra-ST-cells.

The antiviral activity of Shigyaku-to (TJS-109), a traditional Chinese herbal medicine, was investigated in mice infected with herpes simplex virus type 1 (HSV-1). TJS-109 is a combination of the medicinal plant extracts from Zingiberis siccatum rhizoma, Aconiti tuber and Glycyrrhizae radix in a specific proportion. Mice infected with a 10 LD50 dose of HSV-1 were treated with TJS-109 orally at doses of 1. 25 to 20 mg/kg 2 days before, and 1 and 4 days after the infection. The treated groups had 80% (1. 25 mg/kg), 40% (5 mg/kg) and 23% (20 mg/kg) mortality rates 25 days after the infection as compared with a 100% mortality rate in control mice treated with saline. When HSV-1 infected mice (recipients) received CD8+ T cell fractions derived from spleens of mice treated with TJS-109 (donors), 70% of recipients survived, as compared with 0% survivors in the groups of mice treated with saline, B cell fractions, CD4+ T cell fractions or macrophage-enriched fractions prepared from the same donors. TJS-109 did not show any virucidal activities against HSV-1 or any virostatic activities on the growth of HSV-1 in Vero cells. These results suggest that TJS-109 protected mice exposed to lethal amounts of HSV-1 through the activation of CD8+ T cells.

One hundred forty-two kinds of traditional medicines, which have been historically used in China, Indonesia, and Japan, were examined for the antiviral activity of their hot water (HW) extracts against herpes simplex virus type 1 (HSV-1), poliovirus type 1, and measles virus by plaque reduction assay. Thirty-two, 55, and 30 HW-extracts of them showed anti-HSV-1, antipoliovirus, and anti-measles virus activities, respectively. Among the 32 HW-extracts with anti-HSV-1 activity, 3 HW-extracts had anti-HSV-1 activity alone and the others showed anti-HSV-1 activity with anti-poliovirus and/or anti-measles virus activities. The 32 HW-extracts were further examined for their therapeutic efficacies of HSV-1 infection in mice. The mice were infected cutaneously with HSV-1 and HW-extracts were orally administered three times daily. Twelve HW-extracts, currently used for the treatment of various diseases other than viral infection, were found to be significantly effective in limiting the development of skin lesions and/or in prolonging the mean survival times of HSV-1-infected mice. These results suggested that 12 of 142 HW-extracts that exhibited therapeutic efficacy in an animal infection model were possible candidates for anti-HSV-1 traditional medicine.

Either the three-edged needle or the syringe needle, which has been sterilized in 75% alcohol, is used to prick perpendicularly the helix bilaterally, about 0.1 cm deep, and squeeze out 5 drops of blood with hand. Scratch the epiderm of the crus helicis at one side with the knife and embed a mung bean sized medicinal paste, and then fix it with the adhesive plaster. Treat every other three or four days.10 treatments constitute one course. After 7-10 treatments, molt of the cases were cured. After PHA intradermal test, the immune function has been proved increased.
